چکیده

Using an in vitro system for tail attachment, it is demonstrated that following ultraviolet (UV) induction of Salmonella typhimurium LT2 (P22) most of the particles formed lack a functional tail. This effect is found at all temperatures studied, but is more pronounced at temperatures of 37” and above. Similar results are described for thermal and spontaneous induction. Tailless particles are found following infection only at temperatures of 42” and above. Temperature pulse experiments on UV-induced lysogens suggest that late in the growth cycle tail synthesis is inhibited by high temperature. There is also an earlier temperature-sensitive step, which probably causes the production of inactive heads at 40”.

A critical test of the recombination theory of multiplicity reactivation.

Bacteriophage particles are called active when they are able to give rise to production of active phage following adsorption on their specific host bacterium. This is generally detected by the formation of a plaque when plated on nutrient agar by standard techniques (Adams, 1950). If the particles are exposed to ultraviolet light, a fraction of them loses the ability to give rise to production ...

Defective Phage Formation by Lysogens of Integration

Integration deficient (L) mutants of phage P22 can be complemented with L+ phage to yield L mutant lysogens. Once established the L prophage is stable in the absence of L+ function. Ultraviolet or thermal induction of L lysogens leads to product, ion of defective phage particles and low yields of infectious phage. The defective particles contain a normal amount of DNA but the DNA is mainly bacte...

Triple tandem mimotope peptide of Epidermal Growth Factor Receptor displaying on the surface of M13 phage induces anti-tumor response in mice tumor model

Introduction: Epidermal growth factor receptor (EGFR) has been shown to play a critical role in tumor cell growth and its overexpression has been observed in many epithelial tumors. In the field of cancer vaccine research, displaying the peptide mimotope on the surface of phage particles has shown promising results. Methods: In this study using m13-PVIII phage display system, two constructs we...

Bacteriophage control of Shiga toxin 1 production and release by Escherichia coli.

The stx genes of many Shiga toxin-encoding Escherichia coli (STEC) strains are encoded by prophages of the lambda bacteriophage family. In the genome of the Stx1-encoding phage H-19B, the stx(1)AB genes are found approximately 1 kb downstream of the late phage promoter, p(R)', but are known to be regulated by the associated iron-regulated promoter, p(Stx1). Growth of H-19B lysogens in low iron ...

Bacteriophage P22 virion protein which performs an essential early function. I. Analysis of 16-ts mutants.

The product of gene 16 of phage P22, P16, is a head protein. P16 does not play an essential role in phage assembly since particles formed without this protein appear normal by electron microscopy examination (Botstein et al., 1973). P16 is essential when the particle infects a cell in the following cycle of infection (Botstein et al., 1973; King et al., 1973). We have characterized a mutant of ...
